Four colonies of the stingless bee Partamona cupira (Hymenoptera: Apidae) were cytogenetically analyzed using conventional staining and the fluorochromes CMA3 e DAPI. The females have 2n = 34 chromosomes (2K=32<img border=0 src="../../../../../../../img/revistas/gmb/2010nahead/ah06a04CarM.gif" align=absmiddle>+2<img border=0 src="../../../../../../../img/revistas/gmb/2010nahead/ah06a04CarA.gif" align=absmiddle>). Some females, however, presented an additional large B acrocentric chromosome, to a total of 2n = 35. ABSTRACT At present, there is no standardized marker that is routinely used in clinical laboratories to diagnose coccidioidomycosis. Rice blast is the most important fungal disease of rice and is caused by Pyricularia oryzae Sacc. (Telomorph Magnoporthe grisea Barr.). Seven randomly amplified polymorphic DNA (RAPD) markers OPA5, OPG17, OPG18, OPG19, OPF9, OPF17 and OPF19 showed very clear polymorphism in resistant cultivar lines which differed from susceptible lines. By comparing different susceptible lines, nine DNA amplifications of seven primers (OPA5(1000), OPA5(1200,) OPG17(700), OPG18(850), OPG19(500), OPG19(600), OPF9(600), OPF17(1200) and OPF19(600)) were identified as dominant markers for the blast resistant gene in resistant cultivar lines. O objetivo deste trabalho foi validar marcadores moleculares, previamente identificados como ligados ao sexo do mamoeiro, para utilização na seleção indireta em genótipos comerciais. Foram analisadas duas variedades do grupo Solo e dois híbridos do grupo Formosa, com utilização de 20 plantas por genótipo, quatro marcadores do tipo SCAR (Sequence Characterized Amplified Region) e um RAPD (Random Amplified Polymorphic DNA). The soybean (Glycine max (L.) Merrill) frogeye leaf spot is caused by the fungus Cercospora sojina Hara and is a widespread disease in Brazil and other countries, causing severe losses in grain yield and also affecting seed quality. The availability of DNA markers linked to genes for resistance to this disease would accelerate breeding programs, particularly when other traits are also being evaluated. Bulked segregant analysis was applied to 3 F2 populations derived from crosses between the resistant cultivars Parana, Cristalina and Uberaba, and the susceptible cultivar Bossier. Na obtenção de cultivares de feijão com resistência à antracnose, outros atributos agronômicos também devem ser considerados para atender a preferência do consumidor e do produtor. Neste trabalho, objetivou-se identificar famílias de feijão que reúnam, além da resistência à antracnose, alta produtividade, grãos tipo Carioca e porte ereto. Foram cruzados os genitores CNFC 10706, B1 portador do alelo de resistência à antracnose Co-4 e H147 portador do alelo de resistência Co-5. Os três possuem grãos semelhantes ao Carioca. Sequence-Characterized Amplified Region (SCAR) appears as a useful technique for genetic purity testing and variety discrimination, applicable to species in which some other techniques have failed. In particular, this technique is very attractive with species in which RAPD results were not consistent. The RAPD polymorphic bands were cloned, sequenced and from the sequence information, primers pairs for normal PCR were developed. Amplified fragment length polymorphism (AFLP) is a fast and reliable tool to generate a large number of DNA markers. In two unrelated F2 populations of carrot (Daucus carota L.), Brasilia x HCM and B493 x QAL (wild carrot), it was hypothesized that DNA 1) digested with the same restriction endonuclease enzymes and amplified with the same primer combination and 2) sharing the same position in polyacrylamide gels should be conserved sequences. Cinco marcadores STS e SCAR, identificados como associados aos alelos de resistência dos genes Lr1, Lr9, Lr10 e Lr24, foram avaliados por PCR, em 25 genótipos de trigo com conhecida presença ou ausência desses alelos.
